黃芩多醣體之理化特性與抗癌活性

黃芩(Scutellaria baicalensis Georgi)為唇形科植物,廣泛的分佈於中國,其根部為主要研究之對象,近幾十年來,黃芩萃取物及其活性之研究主要是集中在黃酮類等小分子上,較少人去關注黃芩多醣其大分子之研究。本研究目的乃是對黃芩進行不同溶劑萃取得到各多醣體劃分(SB)並進行抗氧化能力之測定以及其理化特性之研究。基本成分結果顯示黃芩含有高量的碳水化合物42.75%以及較多的粗纖維15.48%,脂肪酸組成分析中發現黃芩含有較高的不飽和脂肪酸77.50%以及少量的飽和脂肪酸22.50%,於不飽和脂肪酸中其具有非常高的多元不飽和脂肪酸含量為73.29%以及4.21%的單元不飽和

脂肪酸,當中又以亞麻油酸最多為59.90%。本篇研究運用不同溶劑所製備得之五個黃芩多醣體劃分,以熱水萃後5倍酒精沉澱的SB-1含量最多,其收率達5.51%。多醣體劃分中碳水化合物以SB-3含量最多為78.89%;胜肽的部分則是以SB-2含量最多為60.59%,表示SB-2劃分中不只含有多醣也含有較高量的胜肽。經由單醣組成分析結果表明,五個多醣體劃分中主要都是以木糖為主,其次為葡萄糖還有少量的半乳糖、阿拉伯糖、甘露糖及肌醇所組成。因此從黃芩中萃取得到的五個多醣體劃分主要構造為葡萄木聚糖(glucoxylan)。抗氧化能力的部分由大至小分別為:DPPH自由基清除能力為SB-1>SB-2>SB-3>

SB-4>SB-5;螯合亞鐵能力為SB-4>SB-2>SB-3>SB-5>SB-1;超氧自由基清除能力為SB-3>SB-4>SB-2>SB-5>SB-1;抑制羥自由基生成為SB-1>SB-3>SB-5>SB- 4>SB-2;一氧化氮清除測定為SB-1>SB-5>SB-2>SB-3>SB-4。透過IC50計算結果顯示,SB-1、SB-2具有相較其他劃分最好的抗氧化效果,後續進行傅立葉紅外線分析其官能基,發現SB-1、SB-2是吡喃糖的形式組成並且SB-2是帶有胺基酸訊號的。因此SB-1及SB-2多醣體是很有潛力的,此外進行巨噬細胞存活率發現SB-1在濃度0.2 mg/mL以下;SB-2濃度在0.
