中文摘要由Affymetrix microarray 分析得知TOPK/PBK (T-LAK originated protein kinase/ PDZ-binding kinase)為一個在台灣女性肺腺癌中高度表現的基因。為探討其功能及其與肺癌形成的關聯性,我首先建立了TOPK酵母菌雙合雜交系統,期找出可與TOPK結合的蛋白質,以進一步得知TOPK的生理功能。在本論文中,我找到5個可能會與TOPK結合的蛋白質,包括MCRS1、TRIM37、PPP2R5E、RNF138及RPL12。為驗證上述的蛋白質確實可與TOPK結合,我首先建構了完整的MCRS1與TRIM37基因,並在酵母菌雙合雜交實

驗中證實二者確實可與TOPK結合。此外,TOPK磷酸酶活性喪失的突變(kinase dead mutant),TOPK (AA),可與MCRS1及TRIM37蛋白質有更強的結合。然而,以免疫螢光染色可見TRIM37與TOPK有相同的subcellular location,而MCRS1則否,且只有TRIM37經由co-immunoprecipitation實驗,證實其在細胞內確實可與TOPK結合。然而,TRIM37與TOPK之間的結合,與肺癌形成有何關聯,仍待進ㄧ步的釐清。另外,為直接得知TOPK是否具促進細胞走向癌化的功能,我利用siRNA及shRNA抑制H1299, CL1-0 及具高度移

行能力的 CL1-5 等肺腺癌細胞株內TOPK基因之表現。有趣的是:抑制TOPK基因之表現可有效抑制H1299, CL1-0 及 CL1-5 等細胞之移行及侵入能力;由細胞存活率的分析中也說明上述細胞之移行及侵入能力的抑制並非RNAi(RNA干擾)的副作用所導致。此外,若過量表現TOPK基因於CL1-0 細胞中亦可促進其移行及侵入,顯示TOPK確實可調控細胞的移行及侵入。這些實驗結果指出TOPK在肺癌形成過程中確實扮演著某重要角色。
